Abstract
Optical microscopy is a three-dimensional analysis technique. Historically, an important part of the technological developments in microscopy has been aimed at improving the quality and resolution of the images obtained with this kind of instrument. Due to these improvements emerge several types of optical microscopy, among which the wide-field fluorescence microscope stands out, in which both the image formation and treatment have been the main subject of this thesis. According to this, it can be said that this work applies to the field computational imaging. In this sense, the main contribution made in this thesis, in the framework of research, is the development and testing of model-based algorithm for both, the estimation of parameters of the point spread function and the image restoration in this type of instruments. More generally, with this research work is intended to contribute to the methodology for image processing and analysis, with the purpose that they are adequately prepared for morphometric and photometric quantitative analysis.
